Should "Service Engine Soon" Indictor Turn Off On Its Own?

My Service Engine Soon light was on recently, displaying a code from a problem that was resolved several months ago. The code was read ("Cylinder 1 Misfire") and cleared.  About 5 minutes later, the light came back on.  Today, the light is OFF, without having the code cleared.

Should the Service Engine Soon light turn off on its own, or is this indicating a possible problem with the computer chip?

Also, where is the chip located in a 2001 V6? Is it something that I can replace myself?
